چکیده

27 Diagnosis of Post Kala-azar dermal leishmaniasis (PKDL), particularly the macular form is 28 difficult by microscopy. This study compared the positivity of nested polymerase chain reaction 29 (91.9%) with imprint smear microscopy (70.9%) in 62 PKDL samples. Nested PCR, showing 87.5% 30 positivity in macular lesions compared to 41.6% by imprint smear microscopy, is an efficient method 31 for early diagnosis of PKDL. 32 33 Running Title: PCR vs microscopy in PKDL diagnosis. 34

Comparative evaluation of PCR and imprint smear microscopy analyses of skin biopsy specimens in diagnosis of macular, papular, and mixed papulo-nodular lesions of post-kala-azar dermal leishmaniasis.

Diagnosis of post-kala-azar dermal leishmaniasis (PKDL), particularly the macular form, is difficult when based on microscopy. This study compared the results of nested PCR (91.9% positive samples) with imprint smear microscopy (70.9% positive samples) for 62 PKDL samples. We found that nested PCR, which indicated 87.5% positivity for the macular lesions, compared to 41.6% positivity by imprint...

Evaluation of Polymerase Chain Reaction (PCR) with Slit Skin Smear Examination (SSS) to Confirm Clinical Diagnosis of Leprosy in Eastern Nepal

BACKGROUND Detection of Mycobacterium leprae in slit skin smear (SSS) is a gold standard technique for the leprosy diagnosis. Over recent years, molecular diagnosis by using PCR has been increasingly used as an alternative for its diagnosis due to its higher sensitivity. This study was carried out for comparative evaluation of PCR and SSS microscopy in a cohort of new leprosy cases diagnosed in...

Comparison of fine needle aspiration and direct skin smear in the diagnosis of acute cutaneous leishmaniasis: polymerase chain reaction as a reference method

Background: Accurate diagnosis of cutaneous leishmaniasis wouldavoid unnecessary treatment and scar formation. Direct smear isthe most common method for the diagnosis of this disease but itsnegative result could not rule out the infection; so, the need formore sensitive methods is obvious. We conducted this study tocompare the efficiency of direct skin smear with smears preparedby fine needle a...
